To find the correlation of Glis3 gene polymorphism and diabetic retinopathy (DR) in type 2 Diabetic mellitus (DM) in Northeast China.

• The Northeast Han patients diagnosed as type 2 DM over 7 years by WHO(1999) standard without cardiocerebral vascular diseases and Han health volunteer were included. The fast glucose, visual acuity, intraocular pressure (IOP), indirect ophthalmoscope, slit lamp microscope, ophthalmoscope and fundus fluorescein angiography (FFA) were examined. • All cases were divided into DR group (50 cases), type 2 DM without DR (DNR) group (50 cases )and health control (NC) group ( 50 cases). We analyzed the Glis3 gene polymorphism, 2 sites were involved: rs7041847 and rs7034200. The differences of Glis3 expression and correlation with DR were observed.
The frequencies of AA, AC and CC genotypes were 46%, 30%, 24% in DR group; 48%, 28%, 24% in DNR group; 78%, 14%, 8% in NC group at rs7034200 site. There has significant differences between DR group and NC group (χ²=11.04, p<0.01), DNR group and NC group(χ²=9.90, p<0.01). There has no differences between DR group and DNR group (χ²=0.06, p>0.05). Glis3 allele frequencies at rs7034200 site, there has significant differences between DR group and NC group (χ²=14.61, p<0.01), DNR group and NC group (χ²= 13.58, p<0.01). There has no differences between DR group and DNR group(χ²=0.02, p>0.05). C allele frequencies was higher in NR group and DNR group than NC. After adjusting the risk factors, Logistic regression was used to analyze. It shows that AG+GG and AC+CC were the risk factors of DR or T2DM at rs7041847 or rs7034200 sites.
There are positive correlations between T2DM and Glis3 gene at rs7041847 or rs7034200 site. G and C alleles are heredity risk factor of T2DM in Northeast China . Glis3 was a risk factor of DR, but it did not support that Glis3 induced T2DM progressed to DR.
