To investigate antiproliferative and apoptotic action of aflibercept on fibroblast-like cell strain L929 in vitro.
To study cellular effects of aflibercept on fibroblast-like cells as a model of CNV’s fibroblasts in vitro. To reveal dependance of aflibercept dose on cellular proliferation.
Cultivation of aflibercept with fibroblast-like cell strain L929 was performed in vitro in concentrations of 0.04, 0.08, 0.2, 0.4, 0.5 mcg/ml in 24 hours after cell planting according to standard protocol. Cellular vital indices (survival, mitotic activity, polycaryocyte index) were evaluated microscopically starting from 1st till 5 day of cultivation. Level of apoptosis was determined on the same cellular culture during 5 days. The results were collated to control. Analysis of cellular growth kinetic was performed to asses dose dependence of aflibercept on cellular effects.
After cultivation with aflibercept fibroblast-like cells’ morphology did not undergo significant changes. Cellular density was gradually decreasing starting from the lowest dose. In concentration of 10 and 100 mcg/ml of aflibercept there was complete absence of polykaryotsytes. Mitotic index remained almost stable with slight decrease. Aflibercept did increase the level of apoptosis which had linear dependence to elevated drug concentration. Growth kinetics analysis has reveal marked reduction in cell number. Cellular proliferation rate was constant but cellular density has decreased predominantly by apoptosis. At concentration 200 mcg/ml cellular growth kept decreasing.
Cultivation of aflibercept with fibroblast-like cell strain has revealed antiproliferative and apoptotic effects that were manifested in cellular survival decrease and slight mitosis downregulation starting from the lowest dose. Apoptosis level was elevated and did increase with dose elevation. Antiprolifeartive and apoptotic effects of aflibercept on fibroblast-like cell strain may explain clinical action of the drug on CNV activity and dimensions.
